Job Description
About Stoke
Stoke Therapeutics (Nasdaq: STOK), is a biotechnology company dedicated to restoring protein expression by harnessing the body’s potential with RNA medicine.
Using Stoke’s proprietary TANGO (Targeted Augmentation of Nuclear Gene Output) approach, Stoke is developing antisense oligonucleotides (ASOs) to selectively restore naturally-occurring protein levels.
Stoke’s first medicine in development, zorevunersen, has demonstrated the potential for disease modification in patients with Dravet syndrome and is currently being evaluated in a Phase 3 study.
Stoke’s initial focus are diseases of the central nervous system and the eye that are caused by a loss of ~50% of normal protein levels (haploinsufficiency). Proof of concept has been demonstrated in other organs, tissues, and systems, supporting broad potential for Stoke’s proprietary approach.
Stoke is headquartered in Bedford, Massachusetts. For more information, visit stoketherapeutics.com
Position Purpose
The Regional Medical Director, Value and Access is a senior, field-based Medical Affairs leader who develops and executes the regional medical value strategy and serves as a scientific and strategic partner to payers, pharmacy benefit managers (PBMs), integrated delivery networks (IDNs), accountable care organizations (ACOs), Medicaid stakeholders, health technology assessment (HTA) bodies, and other population-health decision-makers. The role leads unbiased scientific exchange, translates clinical, health-economic, and real-world evidence into decision-relevant value narratives, and shapes regional priorities that support formulary evaluation, coverage, reimbursement readiness, and patient access.
Operating with substantial autonomy, the Regional Medical Value Director partners closely with Field Medical, Market Access, HEOR/RWE, Clinical Development, Regulatory, and Policy.
The role is especially critical in pre-launch, launch, and lifecycle management, where it builds scientific credibility with priority payers and anticipates their evidence needs.
Key Responsibilities
Scientific Engagement
Develop and execute a regional stakeholder engagement plan aligned with medical, evidence, and corporate priorities.
Build peer-to-peer scientific relationships with senior clinical and population-health leaders across national and regional health plans, PBMs, Medicaid organizations, IDNs, ACOs, employer coalitions, and HTA bodies.
Serve as the senior scientific contact for complex payer and reimbursement questions, including pre-launch and launch engagements.
Lead balanced, non-promotional exchange on disease state, clinical data, guidelines, safety and efficacy, comparative evidence, HEOR, RWE, and pipeline information where appropriate.
- Plan and run payer advisory boards and scientific forums, and ensure insights are captured and acted on.
- Represent Medical Affairs at regional and national payer meetings and external forums.
Evidence Communication
Lead regional communication of the totality of evidence: efficacy, safety, comparative data, HEOR, quality of life, budget impact, epidemiology, and RWE.
Develop and maintain payer-facing scientific materials, and evidence summaries through appropriate review processes.
Translate complex evidence into clear, decision-relevant scientific narratives for sophisticated payer audiences.
Keep resources current, rigorous, and aligned with stakeholder needs.
Medical Strategy & Insights
Own the regional medical value plan and contribute to annual and lifecycle planning, prioritization, and resource allocation.
Gather and synthesize actionable insights on unmet needs, evidence expectations, coverage and policy changes, treatment paradigms, and competitive dynamics.
Escalate regional trends and strategic implications to Medical Affairs leadership and cross-functional governance.
Translate insights into recommendations for medical strategy, evidence generation, clinical development, publications, market access, and education.
- Define and monitor regional Medical objectives, KPIs, and impact; adjust plans as needed.
- Contribute senior scientific judgment to portfolio, indication, and lifecycle decisions while preserving Medical Affairs independence.
Cross-Functional Leadership & Capability Building
Provide regional leadership and strategic counsel across Medical Affairs, Field Market Access, HEOR/RWE, Clinical Development, Commercial (within compliance guidelines), Government Affairs, Patient Services, Regulatory, Policy, and Medical Information.
Lead cross-functional workstreams for launch planning, payer evidence planning, advisory boards, congress strategy, and medical education.
Influence priorities and resolve competing needs without direct authority.
Coach and mentor colleagues; Senior Director incumbents may also lead teams, vendors, budgets, or enterprise initiatives.
Inform regional resource allocation, manage assigned project budgets and external partners, and represent the regional perspective in US and global planning forums.
Evidence Generation
Identify and prioritize payer-relevant evidence gaps.
Partner with HEOR/RWE, Epidemiology, Clinical Development, and Market Access on evidence plans, research questions, endpoints, and dissemination.
Initiate or lead rigorous payer research collaborations, pragmatic pilots, data partnerships, investigator-sponsored research, and RWE projects.
Provide senior field input into protocol concepts, publication strategy, congress communications, and medical information resources.
- Evaluate emerging evidence and recommend changes to regional strategy and engagement.
- Governance, Compliance & Operational Excellence
- Operate in accordance with all applicable laws, industry codes, company policies, and SOPs.
Maintain clear separation between scientific exchange and promotion, and apply sound judgment in novel situations.
Document and manage insights, engagements, research collaborations, materials, and transfers of value appropriately.
Identify operational risks, drive continuous improvement, and contribute to scalable field medical value processes.
Complete required training and reporting accurately and on time.
Required Skills & Experience
Advanced clinical or scientific degree in a relevant discipline.
10+ years of progressive experience in Medical Affairs, payer medical engagement, Market Access, HEOR/RWE, clinical development, managed care, or a related pharma/biotech function.
Formal training or demonstrated expertise in health economics, outcomes research, population health, managed care, epidemiology, or health policy strongly preferred.
Skills
Deep expertise in managed care, formulary and coverage decision-making, evidence-based medicine, clinical trial methodology, HEOR, outcomes research, epidemiology, and real-world evidence.
Strategic ability to connect external stakeholder needs with portfolio, evidence-generation, access, and medical priorities.

Ability to critically appraise complex clinical and economic data, identify limitations, and articulate implications for population-health decisions.
Strong business and financial acumen, including prioritization, resource allocation, project planning, and measurement of impact.
Proven influence in matrixed organizations and ability to lead across functions, geographies, and levels without direct authority.
- Sound judgment in ambiguous, high-visibility, and compliance-sensitive situations.
